Title: [abstract] THE EFFECTS OF INCREASED PRESSURES OF HELIUM AND THE NARCOTIC GASES NITROGEN AND ARGON ON CALCIUM UPTAKE BY ACTIVATED NEUROBLASTOMA CELLS.

Abstract: Cultured human neuroblastoma SK-N-SH cells were loaded with the intracellular fluorescent Ca2+ indicator fura-2 AM and either stimulated with carbachol (the cells express M3 type muscarinic receptors) or depolarized with 50 mM KCl. Both evoked a rapid, marked increase in free cytosolic Ca2+ [Ca2+]i determined by fluorescence spectroscopy. Studies in Ca2+-free medium indicated that about 65% of this increase was from uptake of extracellular Ca2+. Non-narcotic He, 9-36 ATA, significantly increased evoked peak [Ca2+]i in response to carbachol or KC1 in a pressure-dependent fashion; eg with carbachol, mean +- sem was 378.8 +- 19.6 for 1 ATA control vs. 481.7 nM +- 24.4 for 36 ATA He (p<.003 by ANOVA). At the same pressures neither N2, nor Ar which is about 2x as narcotic as N2, changed [Ca2+]i significantly from control values, thus apparently negating the pressure effect represented by He. With Ar a slight but not statistically-significant decline from control was seen eg 368.2 +- 31.1 for control, 344.8 +- 16.7 for 36 ATA Ar. Since a rise in intracellular Ca2+ is associated with neuronal activation, these results suggest that changes in Ca2+ fluxes may underly HPNS in divers and its antagonism by narcotic gases.
