Title: [abstract] TISSUE KINETICS OF XENON-133 IN THE DOG KNEE AND GASTROCNEMIUS AT SURFACE PRESSURE.

Abstract: Eight mongrel male dogs weighing between 12 and 25 kg were anesthetized, intubated and placed on a closed circuit ventilator. Shielded cadmium telluride detector probes were positioned over the inspired gas hoses the anterior knee, and the belly of the gastrocnemius. muscle. After stabilization of cardiorespiratory parameters, a bolus of Xe-133 was injected into the respirator circuit. The inlet hose activity level was then maintained constant for 1 h by repeated small injections. A 4 h elimination phase followed with the animal breathing room air. Each animal was studied at rest on two separate days. The tissue activity curves were analyzed using a nonlinear least-squares fit to the sum of 2 exponentials or to an optimum number of exponential terms. The two exponent model for the calf muscle yielded a fast component time constant (TC1) of 18 ± 7 min and a relative weight (W1) of .67 ± .13 and a slow component TC2 of 95 ± 25 min and W2 of .33 ± .13. The anterior knee analysis yielded: TC1 25 ± 17 min, TC2 199 ± 75 min, WI .56 ± 0.14, and W2 .44 ± .14. A residence time analysis of each curve was performed using the fit with the largest number of exponentials in which the parameters could still be estimated with a standard deviation of ± 15%. This analysis produced an average mean tissue residence time (MRT) for Xe-133 in the calf muscle of 37 ± 15 min and in the anterior knee of 101 ± 55 min. Despite tight control of the physiologic parameters, there was a significant day-to-day variation seen in an individual animal's paired MRT's. The estimated standard deviations of the MRT's were 12.6 min for the gastrocnemius muscle and 30 min for the anterior knee. The cause of this variability is uncertain.
