Title: [abstract] HBO2 AFTER HYPOXIA ISCHEMIA IN NEONATAL RAT PUPS

Abstract: INTRODUCTION: Hyperbaric Oxygen (HBO2) has been reported to protect the brain when administered either before or after hypoxic and ischemic insults. However, the potential role of HBO as a neuroprotective treatment is still unclear. In light of this uncertainty we elected to investigate the putative protective effect of HBO2 after brain injury. METHODS: The Rice/Vannucci hypoxia/ischemia model was used to test post injury HBO2 neuroprotection. Wistar rat pups were randomized to: 1) control, 2) positive control from hypoxia preconditioning (HIPC), 3) 3 ata HBO2 for 1 hour at 36C or 4) 3 ata HBO2 for 1 hour at 24C. The rat pups underwent right carotid cauterization (halothane anesthesia) followed by 150 minutes of hypoxia using 8percent O2 (National Welders, National Specialty Gases, Raleigh, NC) in 450 cc 36C temperature controlled plexiglas chambers. One hour after the hypoxia the experimental pups received a 1 hour "rescue treatment" of HBO2 at the temperatures and pressures stated above. All compressions and decompressions were conducted at a target travel rate of 0.25 ata per minute to avoid compression related temperature changes. On post-natal day 14 the forebrain was sectioned at the midline and the ipsilateral hemisphere decrement was calculated. Parametric intergroup comparisons were made using a one-way ANOVA with post hoc Tukey's HSD and confirmed by non-parametric Kruskal-Wallis H using SPSS v 8.0. P values of 0.05 were considered significant. RESULTS: The positive controls confirmed the neuroprotective effect of HIPC as reported previously ( p=.02 Kruskal-Wallis). However, no neuroprotective effect for HBO2 either at 37C or 24C was observed.
