Abstract: BACKGROUND: Pressure has been shown to antagonize the behavioral effects of anesthetics and sedatives in most organisms. Recent evidence suggests that this is not due to displacement of drug from its binding site, but rather a specific excitatory effect which functionally competes with anesthetic depression. In an attempt to find organisms that lack this excitatory pathway, we examined dose-response relationships for ethanol in the nematode C.elegans at 1 and 100 ATA. METHODS: Wild-type (N2) nematodes in M9 buffer with 0-1000 mM ethanol were exposed to either 1 or 100 ATA in a 4 ml hydraulically compressed chamber. After 20 min., -1000 worms 1.5 were scored as either not moving (0), moving slowly (1) or moving normally (2). The fraction in each category was multiplied by the score and summed to yield an movement score ranging from 0 to 2. RESULTS: 100 ATA pressure alone depressed movement slightly (reversible with return to 1 ATA). This small depressant effect of pressure persisted throughout the ethanol dose-response curve (fig 1). The EC50 for ethanol at 100 ATA (363±16; mM±SEM) is lower than that at 1 ATA (478±44)(P<0.01). CONCLUSIONS: This is further evidence against pressure displacement of anesthetic molecules from sites of action since this premise requires any organism responding to anesthetics to show pressure reversal. Pressure reversal has also not been observed in a few organisms which apparently lack glycine neurotransmission, suggesting that pressure-antagonism of this inhibitory pathway produces the excitation necessary for competition with the anesthetic, and for symptoms of HPNS. We speculate that C. elegans also lacks such neurotransmission. (funded by GM-46724). <graph>
